The PetakaG3 is a cell culture device designed to culture adherent and non-adherent cells. The device eliminates all air space inside the culture chamber to allow the anchorage and growth of adherent cells on all internal surfaces.



The PetakaG3 is especially designed to grow cells in an isolated, auto-modified milieu, that is highly protected from external environmental conditions. That protection permits effective cell incubation with or without external  CO2 control and independent of relative humidity in the atmosphere. A proprietary structural design establishes a gas exchange rate that provides a suitable growing environment for the majority of cells incubated in regular atmosphere (0.2% CO2). Additionally, this structure reduces dehydration to virtually zero which allows the storage of media preloaded devices for weeks. Therefore the PetakaG3 can be effectively incubated in any temperature-controlled chamber without CO2 and humidity controlled environment.

The internal cell culture chamber has 150 cm2 of surface area available for cell attachment and growth and 20 ml (±1%) of volume. This optimized media/surface area rate (0.133 ml/cm2) facilitates the growth of large numbers of cells per PetakaG3 (the number is dependent upon the cellular metabolic and doubling time characteristics of the cells) without media replacement.  This allows for optimal use of media and special additives such as growth factors.

PetakaG3 Versions

There are five versions of the PetakaG3:

- PetakaG3™ LOT (Low Oxygen Transfer
- PetakaG3™ ET™ (Easy Transfer)
- PetakaG3™ HOT (High Oxygen Transfer)
- PetakaG3™ FLAT Suspended Cell Culture)
- PetakaG3™ MATRIX  (Specific Molecular Matrix)

The PetakaG3 HOT, FLAT and MATRIX are manufactured from a co-polymer called Styrolux and are specifically designed for high oxygen respiring cells such as liver cells, monoclonal hybridomas, etc. Cells can be put into dormancy for transportation or short-term storage but the high gas diffusion material of the PetakG3 HOT does not allow the oxygen level to go below 2%; it is therefore unsuitable for long-term dormancy (more than 30 days without media change). Culturing for more than two weeks is also not recommended as the PetakaG3 walls not only allow gas to diffuse through but they also allow minute amounts of water to escape.

The PetakaG3™ FLAT bioreactor has all of the same size and structural characteristics of the PetakaG3™ HOT bioreactor except that the interior surfaces are not treated to promote cell adherence to the walls. The PetakaG3™ MATRIX is also similarly constructed to HOT but in this version the internal surfaces are coated with a specific molecular matrix such as collagen, fibronectin, poly-L-lysine, Matrigel, gelatin and laminin. This product is best suited to growing different types of stem cells.

The PetakaG3 LOT and ET are manufactured from a crystal polystyrene called Styron and are designed for most mammalian cells with a low to medium oxygen diffusion rate such as CHO cells. This is the material from which all previous versions of the Petaka were manufactured. This material allows up to two months of dormancy without media change of low/medium oxygen respiring cells and up to several months with media change. Culturing in PetakaG3 LOT brings the dissolved oxygen levels in the media down to 1.5% that induces dormancy at confluence.

The PetakaG3™ ET™ bioreactor has all of the same basic characteristics as the PetakaG3™ LOT but contains magnetic particles inside the main cell culture chamber.The Petaka™ ET bioreactor is used in conjunction with the ClearCell™ instrument to mechanically detach cells with minimal cell damage. This allows cell harvesting without using enzymes or chemicals such as Trypsin-EDTA and the ET version is well suited for culturing cells with special protocols such as those for stem cell cultures.

The Petaka G3 Personal Incubator has been specially designed to integrate with the PetakaG3 cell culture system. Key capabilities of the Personal Incubator include:

Centrifuge for the PetakaG3



The Digicen 21 has been especially adapted to centrifuge 8 PetakaG3™ simultaneously in made-to-measure sleeves. Key features include:
  • lluminated LCD screen
  • Centrifugation speed up to 20,000 rpm
  • Allows the regulation of rpm in increments of 100 rpm,  in increments of 10 x g, & time setting of 1 to 99 minutes or indefinite time setting

The Petaka storage stand/rack is simple and safe. The stand can hold up to 12 Petaka culture units in a vertical position and two stands can be combined to hold the Petakas in the horizontal position. Features of the Dispensing Tips include:

    The CellMaker PLUS is a state-of-the-art, easy to use, single-use, disposable bioreactor control system. The unit is fully automated in order to speed up and simplify cell culture and fermentation bioprocesses and includes a wealth of new technologies for efficient growth of the widest range of cell types. It is an innovative, high quality, robust system which can be used with bacteria, yeast, algae, bacteriophage as well as the more challenging mammalian and insect cells. Cells are cultured in a unique, single-use, disposable cell culture bag, the CellexusBag™, that expedites set-up times and eliminates the extensive cleaning and sterilizing requirements of other systems.



    CellMaker Plus condenser system and optional condenser liner

    Unlike other disposable systems, which need shakers, rockers, rollers or large environmental chambers, the CellMaker PLUS eliminates mechanical methods for mixing by employing a proprietary airlift technology for optimal mixing and oxygenation of the cell culture. This is highly efficient, quick, simple, scalable and inexpensive to use. It provides optimal aeration along with precise temperature control enabling production of high cell densities and maximizing expression levels of soluble protein. The CellMaker Plus consists of four modules: an enclosure, a controller, a supply hub and an oxygenator:

    Enclosure

    The Enclosure holds the single use CellexusBag and incorporates a Peltier driven thermal plate heater, a built in chiller and a PT100 temperature sensor to enable cell culture temperature to be closely monitored and controlled. Enclosures are available in 8 L and 50 L sizes for use with 1-8 L and 10-50 L CellexusBags in order to provide scale flexibility. A high efficiency condenser system removes moisture from an exhaust line before it is vented through a microbiological air filter and optional gamma-irradiated, single-use condenser liners are available to provide ease of use and reduced set-up/turn-around times. The Enclosure also incorporates transparent panels to allow visualization of culture progress and to enhance performance for light-dependent applications.



    CellMaker Enclosure

    Controller

    Key protocol parameters are entered into the controller via a 15" touchscreen monitor. Control of dissolved oxygen (DO) is essential to achieve optimal cell concentrations and to ensure the viability of mammalian and insect cells and so the Controller is available with an optional pH and oxygen sensor reader (which can be retrofitted) in order to measure pH and dissolved oxygen concentration. Automated feedback of data enables the Controller to adjust the pH of the culture and to regulate air, oxygen and carbon dioxide flow.

    SupplyHub

    The SupplyHub incorporates an air pumping system, temperature regulation as well as process measurement and data acquisition. It also provides a power source to the Controller and Enclosure.

    Oxygenator

    The Provides a continuous supply of oxygen and can dovetail with the SupplyHub or be used as a stand-alone unit.
